Abstract
In this study, the highest spore number was obtained by single factor experiment and box-behnken response to the medium formulation and culture conditions for producing spores of Bacillus coagulans 13002. Finally, the optimal medium formula was determined as glucose 15.0 g, bacteriology peptone 10.0 g, yeast extract 20.0 g, NaCl 10.0 g, MnSO4 0.2 g, K2HPO4 3.0 g, and 10% bran water extract to 1 L. The optimal culture conditions were the initial pH value of 7.0, inoculation amount of 6%(V/V), liquid loading amount of 30 mL(250 mL conical flask), and culture at 200 r/min at 40℃ for 52 h. The final spore number was 3.31×109CFU/mL. This study will lay down a foundation for industrial scale production of Bacillus coagulans 13002.
