Proliferation Effect and Mechanism of Bifidobacterium from Infantis on Human Fetal Colon Epithelial Cells

  • The proliferative effects of fermented supernatants and crushed matter from 12 infant-derived Bifidobacterium on human fetal colon epithelial cells(CCD841 CoN)were analyzed. The bifidobacterial with the best pro-proliferative effect were screened out,and their surface and secreted proteins were extracted. The effects of these two types of proteins on cell proliferation,cell cycle,and proliferation-related genes expression were examined,and the role of CCD841 CoN cell proliferation was explored. The results showed that Bifidobacterium bifidum H3-R2 was screened,and it contained 50 μg/mL of surface protein and 10 μg/mL of secreted protein. After incubation with CCD841 CoN cells for 24 h,it had a significant proliferation promotion effect on the cells(P<0.05)and increased cell S-phase ratio,decreased G0/G1 phase ratio(P<0.05),increased intracellular β-catenin,c-Myc and Cyclin D1 gene mRNA transcription levels(P<0.05),decreased Axin2,GSK-3β gene mRNA transcription level(P<0.05). In summary,infant-derived bifidobacterial could promote the proliferation of human fetal colon epithelial cells at the level of mRNA transcription,and had potential application value in promoting the intestinal development of newborns.
