Differential Expressed Protein Analysis of Aspergillus fumigatus Treated with Propolis Ethanol Extract by iTRAQ

  • To further explore the antifungal mode of propolis against Aspergillus fumigatus,the ethanol extracts of propolis was treated by A.fumigatus. The iTRAQ technology was used to analyze the possible mechanism,and then the functions of proteins were analyzed by bioinformatics. A total of 284 differentially expressed proteins(EDGs)were identified,of which 123(43.5%)displayed to be up-regulated and 161(56.5%)down-regulated. According to GO function analysis of EDGs,proteins were mainly involved in the processes of inorganic ion transport,liposome transport,energy and carbon metabolism,and signal transduction. According to COG functional-categories,the EDGs were divided into 21 functional categories,including energy production and conversion,carbohydrate transport and metabolism,cell cycle control,chromatin structure and dynamics,and so on. The protein with the greatest difference in expression was tubulin,which was up-regulated for 6-fold. Proteomic data showed that propolis could change the expression level of many proteins such as respiratory electron transport chain,ATP generation and cytoskeleton,indicating that propolis could interference energy metabolism and interrupt the cell division to inhibit the growth of A. fumigatus.
