Effect of Chicken Hydrolysate on Antioxidative Activity of HepG2 Cells and Antioxidative Enzyme Activity of Mouse Serum

  • In order to study the antioxidative effects of SSPH (Salt Soluble Protein Hydrolysate) and MFH (Muscle Fibrin Hydrolysate) on HepG2 cells, the activity of ROS in HepG2 cells and the activity of antioxidant enzymes in mice serum after 28 days of gastric gavage were determined. SSPH and MFH were two kinds of enzymatic hydrolysates, which were extracted from chicken breast meat under special conditions and prepared by enzymolysis, with molecular weight more than 95% and less than 1000 u. The results showed that the survival rate of HepG2 cells was 84.50% and 89.87% when SSPH and MFH were added in the concentration of 500 μg/mL. Chicken hydrolysates were not toxic to HepG2 cells. In HepG2 cells experiment, the fluorescence intensity of HepG2 cells in the experimental group was significantly lower than that in the blank control group (P<0.05), which reduced with the increase of chicken enzymatic hydrolysate concentration.When the concentrations of SSPH and MFH were 150 μg/mL, the cellular antioxidant activity (CAA) values were 43.17% and 46.64%, respectively, which indicated that both of them had good scavenging capacity of cellular ROS and showed a good dose-response relationship. In mice experiment, normal saline and 0.2 mL dose of 200 mg/kg·bw·d chicken homogenate were used as control group.The antioxidant enzyme activity of SOD, CAT and GSH PX in mice serum was analyzed after 28 days of gastric gavage and 30 min of swimming.The results showed that the activity of SOD, CAT and GSH-Px in serum were extremely significant (P<0.01) or significantly (P<0.05) higher than normal saline and the same dose chicken homogenate (40 mg/kg·bw·d and 200 mg/kg·bw·d), except the CAT activity of SSPH for 40 mg/kg·bw·d.The results showed that SSPH and MFH had significant effects on the activity of antioxidant enzymes in mice.
