Study on Determination of Mononucleosides in Morchella sextelata Mycelium by High Performance Capillary Electrophoresis

  • A new method using high performance capillary electrophoresis-diode array detector for the analysis of mononucleosides in the extracts from Morchella sextelata was described.It was determined that the dried mycelium was used to extract the sample according to the method that it extracted in 30 times water by Ultrasound first and then followed by water bath.In order to simultaneously determine five mononucleosides (3-deoxyadenosine, adenosine, guanosine, uridine andinosine), the conditions of high performance capillary electrophoresis (HPCE) were optimized.The optimum conditions were determined as follows:50 mmol/L borax buffer with pH9.4 and β-cyclodextrin of 1.0 mg/mL as running buffer, separation voltage of 25 kV, injection time of 8 s.Under the optimum electrophoresis conditions, the five standards were separated in 20 min, and the RSD of migration time was less than 0.89% and the RSD of peak area was less than 2.9%.In the stability test, the RSD of the five standards in the 8 h migration time was less than 0.61%, and the RSD of peak area was less than 4.73%.It indicated that the sample had good stability within 8 h.The results showed that the method could be used to determine the content of 3 deoxyadenosine, adenosine and uridine in Morchella.The content of 3-deoxyadenosine in the mycelium of the morel was 4.38 mg/g, the adenosine content was 6.6 mg/g, and the uridine content was 7.8 mg/g.The recoveries of the three mononucleoside compounds ranged from 95.5% to 100.8%, and the RSD was less than 4.9% (n=3).This method was suitable for the determination of mononucleoside compounds in Morchella sextelata hyphae.
