The Prevention Effect of Elaeagnus mollis Oil on High-fat Diet Induced Obesity in Mice

  • Objective:The aim of this study was to investigate the decreasing effect of Elaeagnus mollis oil(EMO)on hyperlipidemia and obesity induced by high-fat diet. Method:Forty male C57BL/6 mice were randomly assigned into 5 groups(n=8):normal diet group,high-fat diet group and low-,middle- and high-dose EMO groups(gavaged with 1.5,3.0,6.0 g/kg·d of EMO,respectively). The mice in the normal diet group were fed with normal diet,while the remaining groups were fed with high-fat diet for 8 weeks. Body weight and food intake were monitored throughout experiment. At the end of study,the levels of serum TC,TG,HDL-C and hepatic TC,TG were measured. The content of serum LDL-C and AI value were calculated,and histological examinations were conducted to analysis the number and size of adipose tissue cells. Meanwhile,the correlation analysis was performed. Results:The results showed that Elaeagnus mollis oil significantly reduced body weight,fat weight,liver weight,Lee's index,hepatic TG level,serum TG,TC and LDL-C levels compared with HFD group(P<0.05),especially the middle-dose EMO group. The supplement of EMO increased the level of HDL-C and decreased AI value,but no significant change was observed(P>0.05). Moreover,the result of HE staining showed that adipocyte became larger and disordered on account of high-fat diet,while supplementing EMO effectively blocked adipocytosis,and the improving efficiency was more prominent of middle-dose EMO group. Correlation analysis showed that adipocyte hypertrophy displayed a positively correlated with hepatic TG and TC,a negatively correlated with serum HDL-C and a positively correlated with AI(P<0.05),which suggested that Elaeagnus mollis oil could reduce lipid content in liver and regulate lipid metabolism to inhibit adipocytosis,and achieved the aim of reducing hyperlipidemia and obesity. Conclusion:Elaeagnus mollis oil can be beneficial as a functional food to prevent obesity and other obesity-related metabolic diseases.
