Optimization of Polysaccharides Extraction from Seed Melon by Response Surface Methodology and Its Hypolipidemic Effects in Vitro

  • The seed melon planted in Xinjiang was used as the raw material to extract the polysaccharides. The extraction process of seed melon polysaccharides was optimized by response surface methodology(RSM)based on single factor experiments. The optimal extraction process of ultrasonic-assisted extraction of seed melon polysaccharides was as follows:Extraction temperature 42℃,extraction time 2 h,solid-liquid ratio 1:30 g/mL,ultrasonic power 220 W. Under these conditions,the extraction rate of seed melon polysaccharide was 6.03%,which was consistent with the predicted value,indicating that the model was well fitted and the process conditions were accurate and feasible. The binding ability of different concentrations of seed melon polysaccharides with sodium glycocholate and sodium taurocholate was studied. When the concentration of the seed melon polysaccharide was 2.5 mg/mL,the binding rate of sodium glycinate and sodium taurocholate reached 62.67% and 69.27%,respectively,showing that the seed melon polysaccharide had better hypolipidemic activity in vitro. Compared with the results of the cholate-binding test of other extracts,the hypoglycemic effect of the seed melon polysaccharide was more effective. So,the seed melon polysaccharide was a potential natural hypolipidemic active ingredient.
