Differental Proteomic Analysis in Breast and Leg Muscles from Yunnan Yanjin Silky Fowl Chicken and Wuding Chicken

  • In the study,Yanjin silky fowl chicken and Wuding Chicken were chosen to analyze the protein spots expressed differently from the breast muscle and leg muscle by two-dimensional gel electrophoresis(DE). And the peptide mapping fingerprint was obtained by matrix-assisted laser desorption ionization-time of flight-time of flight-mass spectrometry(MALDI-TOF/TOF MS). The results showed that the protein spots were distributed mainly within the range of pH4.0~7.0. For Yanjin silky fowl chicken leg muscle and breast muscle,the average protein spots detected were(1421±34)and(1329±25)spots,respectively. For Wuding Chicken leg muscle and breast muscle,the average protein spots detected were(1585±19)and(1516±28)spots,respectively. There were 178 protein spots expressed differently by analyzing the 2-DE maps between YanJin silky fowl chicken muscle and Wuding Chicken muscle. For Yanjin silky fowl chicken muscle,there were 72 protein spots expressed higher,and for Wuding Chicken muscle,106 protein spots expressed higher. The protein spots were identified as lamin-A,phosphoglucomutase 1,beta-enolase,isoform 1 of tropomyosin alpha-1 chain,ATP synthase subunit beta,C-reactive protein precursor,alpha-crystallin B chain,creatine kinase M-type,glyoxylase 1,phosphoglycerate,slow skeletal muscle troponin T,proteasome non-ATPase regulatory subunit 14,heat shock protein beta-1,superoxide dismutase and some hypothetical proteins and uncharacterized proteins.
