Alleviating Effect of Garcinia Extract on Acute Gouty Arthritis Induced by Sodium Uric Acid in Rats

  • In order to study the alleviating effect of garcinia extract on acute gouty arthritis induced by sodium urate in rats, Sprague Dawley rats were randomly divided into normal group, model group, GE group (300, 200, 100 mg/kg).The rats in each group were divided into groups for six days. Except for the normal group, the rats in the other groups were given sodium urate for modeling. After the model was used to determine the swelling degree of rat joints, gait analysis was performed, and the animal was killed after eosin (HE) staining, the joint cavity effusion smear and synovial infiltration were observed, and the enzyme-linked immunosorbent assay was used to determine the Leukocyte-1β (IL-1β) and interleukin-1α in synovial tissue of rats (IL-1α), tumor necrosis factor-α (TNF-α) content. The results showed that the left ankle of the model group was swollen, the gait was limp, the inflammatory cells infiltrated in synovial tissue, and the inflammatory factor IL-1β, IL-1α, TNF-α were significantly (p<0.05) higher than the normal group, which suggested the success of modeling. The swelling degree of ankle in the high-dose group of garcinia extract improved with time, the gait swelling was also eased, with the inhibition rate of joint swelling in the high-dose group was kept above 50% all the time, and the inhibition rate in the middle-dose group reached 47.1% after 6 h of inflammation. The inflammatory factor IL-1β, IL-1α, TNF-α content were significantly (p<0.05) lower than these in the model group, while the low dosage group of garcinia extract had no significant difference compared with the model group. The above results showed that the high dosage group of garcinia extract have positive effects on acute gouty arthritis induced by sodium urate in rats.
