Optimization of Extraction Process and Protective Effect of Cellular Senescence from Asarum Polysaccharides

Abstract
Objective:To optimize the extraction process of Asarum polysaccharides and evaluate the protective effect of Asarum polysaccharides on senescence of vascular smooth muscle cells. Methods:The Box-Behnken response surface method was used to optimize the extraction parameters of polysaccharides,ÄKTA explorer 100 chromatography system was used to separation and purification of polysaccharides,the protective effect of Asarum polysaccharides on Rat aortic smooth muscle cellular senescence induced by Etoposide was investigated by morphological observation,MTT assay,senescence β-galactosidase staining. Results:The optimal extraction conditions of Asarum polysaccharides were as follows:processing temperature 90 ℃,extraction time of 2.37 h and ratio of water to raw material of 40:1 mL/g,the maximum yield was 10.20%. In cellular experiments,Asarum polysaccharides pretreatment significantly reduced the morphological changes of cellular senescence,improved cell viability,and down-regulated the level of senescence β-galactosidase activities induced by Etoposide. Conclusion:The optimum extraction process of Asarum polysaccharides was determined,Asarum polysaccharides could significantly inhibit senescence and apoptosis of Rat aortic smooth muscle cells induced by Etoposide,which has protective effect on cardiovascular disease.
