Effects of Ganoderma lucidum Extracts on Lipid Metabolism and Gut Microbiota in High-Fat Diet Fed Rats

  • Objective:To study the influence of Ganoderma lucidum extracts(GLE)intervention on gut microbiota and lipid metabolism in rat by hight-fat diet. Methods:Twenty four rats were randomly divided into three groups,including control group(standard normal diet+distilled water),model group(high-fat diet+distilled water,HFD)and GLE group(high-fat diet+150 mg/kg·d GLE,GLE). All treatments were conducted with 2 mL/d by oral administration for 8-week continuous period. After 8 weeks,serum and liver markers and related mRNA expression levels were measured,and the feces for fourth weeks and eighth weeks were collected. Results:Compared with HFD group,oral administration of GLE markedly alleviated the dyslipidemia through decreasing the levels of serum and liver total triglyceride(TG),total cholesterol(TC),low-density lipoprotein cholesterol(LDL-C)(p<0.05),and increasing high-density lipoprotein cholesterol(HDL-C)in liver(p<0.05). In addition,GLE treatment resulted in reductions in mRNA levels of SREBP-1C and ApoB,but elevations in PPARα gene expression(p<0.01). Furthermore,GLE supplementation altered the composition of gut microbiota,especially increased the relative abundance of Phascolsrctobacterium,Coprococcus,Sutterella and Desulfovibrio,and reduced the relative abundance of Anaerotruncus,Dorea,Streptococcus,Lactobacillus and Paraprevotella. Conclusion:GLE could improve the lipid metabolism disorder and intestinal flora imbalance in Wistar rats by high-fat diet.
