Response Surface Optimization of Simultaneous Extraction of Protein and Soluble Dietary Fiber from Grape Seed Meal by Complex Enzymatic Method

  • The protein and soluble dietary fiber of grape seed meal were extracted by complex enzymatic method.Papain and cellulase were used in the experiment to extract. On the basis of single factor experiments, response surface methodology was applied to optimize the extraction conditions. The extraction rate of protein and soluble dietary fiber from grape seed meal was determined by the complex enzyme.The optimum conditions were as follows:the papain and cellulase was added 3:1, the extraction temperature was 50℃, the ratio of material to liquid was 1:20 (g/mL), pH7.0, the extraction time was 50 min. The extraction rate of protein was 83.07%±1.43% and the extraction rate of soluble dietary fiber was 34.04%±0.87%.Therefore, it was feasible to select the synchronous extraction of protein and soluble dietary fiber from grape seed meal by using complex enzymatic method. It also provided theoretical guidance for the synchronous extraction of protein and soluble dietary fiber by other by-products.
