Optimization of Hydrolysis Process of Polysaccharides from Chrysanthemum indicum Flowers by Response Surface Methodology

  • Based on the single factor experiments,the response surface methodology was employed to optimize the hydrolysis process of wild Chrysanthemum polysaccharides. Taking the wild Chrysanthemum as the research object,the influence of hydrolysis time,dosage of trifluoroacetic acid and temperature on the hydrolysis of wild Chrysanthemum polysaccharide was determined by single factor experiment. The total peak area of monosaccharides in the gas chromatogram was used as the response value,and combined with response surface method the optimal hydrolysis conditions for wild Chrysanthemum polysaccharides were determined. The results showed that the optimum hydrolysis conditions of the polysaccharides of Chrysanthemum chrysanthemum were as follows:the temperature at 110℃,the hydrolysis time was 9 h,the amount of trifluoroacetic acid was 3 mL,and the single and total peak area measured by gas chromatography under these conditions was the largest. The model value was predicted to be 4162.26 and the actual value was 4151.35,which was similar to the predicted value. The relative deviation was 0.26%. It showed that the optimized hydrolysis method of the Chrysanthemum polysaccharide obtained by this model was reasonable and the conditions were stable and feasible.
