Ameliorative Effects of Chondroitin Sulfate Disaccharides on the Murine Renal Inflammation Under Fatigue-stressed Conditions

  • Objective:To investigate the ameliorative effects of chondroitin sulfate disaccharides on the murine renal inflammation under fatigue-stressed conditions. Methods:Total 24 Balb/c mice were randomly assigned into four groups:the normal group (N group), fatigue-stressed group (M group), normal mice intervened by chondroitin sulfate disaccharides group (150 mg/kg·bw) (CS group) and fatigue-stressed mice intervened by chondroitin sulfate disaccharides group (150 mg/kg·bw) (S group). The mice in both M and S groups were subjected to a forced exercise wheel-track treadmill, which commenced exercise at 20 r/min running speed for 3 h. Rested for 5 d after 2 d of continuous exercise. Experimental period was 16 d and a total exercise time was 6 d. After interventions, the superoxide dismutase (SOD) activity, the concentration of methane dicarboxylic aldehyde (MDA), the mRNA expression levels of transforming growth factor (TGF-β1), nuclear transcription factor (NF-κB), interleukin-6 (IL-6), and monocyte chemoattractant protein-1 (MCP-1) and the protein expression of NF-κB, IL-6, and MCP-1 in kidney were detected. Results:Compared with the normal group, the activity of SOD decreased, the concentration of MDA, the mRNA expression levels of TGF-β1 and the mRNA and protein expression of NF-κB, IL-6, MCP-1 were significantly increased in mice in M group (p<0.01). The activity of SOD was significantly improved (p<0.01) in CS group. Compared with M group, the activity of SOD was significantly improved (p<0.05) and the concentration of MDA, the mRNA expression of IL-6 and MCP-1 were significantly decreased (p<0.01), and the protein expression of IL-6 and MCP-1 were significantly decreasd (p<0.05) in S group. Conclusion:Chondroitin sulfate disaccharides could improve mice renal inflammation induced by fatigue-stress, the mechanism might be related to the improvement of renal oxidative stress.
