Analysis of gingerol compounds in rhizoma zingiberis by HPLC-ESI-Q-TOF-MS/MS

  • In order to rapidly and accurately analyze the composition of gingerol in rhizoma zingiberis, a detection method of liquid chromatography electrospray ion trap time-of-flight tandem mass spectrometry was established. Syncronis C18 (5 μm×250 mm×4.6 mm)chromatographic column was used to analyse, and a binary solvent system composed of acetonitrile and water containing a 0.1% of formic acid was used as mobile phase, then ginger samples were detected by electrospray ion trap time-of-flight mass spectrometry. The scanning mode is full ion, positive ion and negative ion mode. Compared to standard substance, the compositions of gingerol were analysed. The results showed that 5 kinds of gingerol standard sample set in the linear relationship between the concentration range 0~200 μg/mL (R2 ≥ 0.9992), the recoveries were between 95.48%~99.46%, the repeatability (RSD:0.87%~3.26%)and precision of the instrument (RSD:0.02%~1.24%)were nice. Finally, according to the parent ion and fragment ions of standard substance and references we can analyse gingerol compounds composition of rhizoma zingiberis. After analysis, we identified the precise five gingerol compounds, and compared with the HPLC this method showed good sensitivity and resolution.
