Study on the polysaccharide accumulation of Dendrobium officinale in rapid propagation

  • To obtain fast-breeding and high medicinal-value.D. officinale,polysaccharide content was identifying indicator. Taking PLB as a carrier,Nitrate/ammonium molecule ratio,sucrose and BA concentration were recognized the most important factors in seven medium compositions affecting PLBs polysaccharide content through the Plackett-Burman design. The steepest ascent experiment and further Box-Behnken design response surface methods were designed,optimal results of the presence of regression model were as follows:nitrate/ammonium molecule ratio,sucrose and BA concentration were 1.90:1,29.1 g/L,1.65 mg/L.Under these conditions,the mass fraction of the polysaccharide increased 33.95% with a significant improvement compared with the value of 25.36% before optimization. The optimization effect was remarkable.
