Determination of neohesperidin dihydrochalcone in beverages by reversed phase high performance liquid chromatography

  • A method of reversed phase high performance liquid chromatography was established for the determination of neohesperidin dihydrochalcone in beverages.The extraction method, precipitant and chromatography conditions were optimized.The separation was performed on an Accucore C18 ( 2.6 μm, 4.6 mm × 100 mm) with methanol and water ( 45∶ 55, v/v) isocratic elution as the mobile phase at a flow rate of 1.0 m L/min and 25 ℃.The detection wavelength was at 283 nm and the sample injection volume was 10 μL. Neohesperidin dihydrochalcone was isolated with satisfactory resolution within 10 min. The concentration of neohesperidin dihydrochalcone in the range of 0.5 ~ 50 mg/L was linearly correlated to the peak area, with correlation coefficients higher than 0.999 and detection limit of 0.15 mg/L.The recoveries of neohesperidin dihydrochalcone at three spiked levels ( 10, 40, 80 mg/kg) were in the range of 90.2% 103.5% and the relative standard deviations ( RSDs) were in the range of 1.96% ~ 7.59% ( n = 6) . The method is specific, sensitive, simple and suitable for the determination of neohesperidin dihydrochalcone in beverages.
