Double function yeast cell-wall extract and its capability of adsorbing aflatoxin B1

  • In this study, we managed to produce a type of cell-wall extract with double functions i.e.spore inhibition and toxins adsorption by using thymol to anchor the phospholipid of yeast cells and lyse them. It was showed that thymol led to the yeast cell death with the observation by using the flow cytometry and fluorescence microscope.When yeast cell-wall extract by yeast cell was treated with 500 μg/m L thymol, the content of β-glucan was increased by 51.5% in the cell-wall extracts by phenol-sulfuric, and soluble protein in cells was decreased by 40% by SDS-PAGE.This indicated that thymol gave rise to the lysis of yeast cell and destroyed the integrity of the cell membrane, the structural of peptidoglycan on the cell wall changed, yeast cell released their contentand and the adsorption capacity of AFB1 increased.The capability of cell-wall extract adsorbing AFB1 was 2.5 μg/mg by thin layer chromatography ( TLC) and high performance liquid chromatography ( HPLC) . And its thymol-loading capacity was 41.5 μg/mg. This study provided new ideas and theoretical basis for the control of aflatoxin contamination in food and feed.
