Establishment of optimum media for lipid production in Yarrowia lipolytica by Taguchi method

  • Taguchi orthogonal experimental design( TOED) was used to optimize the fermentation media of lipid production by Y.lipolytica with crude glycerol as carbon source in this study.Based on single factor test,TOED was established by Minitab 17.Then the means and SNRs( Signal to noise ratio) of each factor were analyzed and the results showed that C/N and the concentration of MgSO_4·7H_2O in the medium had a substantial significant impact on lipid production,the CaCl_2 concentration had a significant influence on lipid production. The optimal medium composition was C/N 100 ∶ 1( crude glycerol 50 g/L,ammonium sulfate 1.08 g/L),MgSO_4·7H_2O 3 g/L,CaCl_2 0.2 g/L,KH_2PO_4 4g/L. The fermentation broth with optimal media were incubated at 28 ℃ and 180 r/ min for 4 days,the lipid yield reached 1.859 g/L and the predicted lipid yield of fitted model was 1.782 g/L.The oil yield increased by 48.5% compared with the orginal.
