Content determination of kinsenoside in Jin-Xian-Lian from three Anoectochilus species by HPLC-ELSD

  • To establish a HPLC- ELSD method for determination of kinsenoside in Jin- Xian- Lian from three Anoectochilus species,the HPLC system consisting of phenomenex NH2column( 250 mm × 4.6 mm,5 μm) and an isocratic elution system of acetonitrile- water( 92∶ 8,V / V) as the mobile phase was adopted.The flow rate was set at 0.8 m L / min,the column temperature was room temperature,ELSD spray chamber temperature was 70 ℃,and the nitrogen flow rate was 1.5 L / min. The results indicated that the liner range of kinsenoside was 76.02 ~ 2432.64 μg / m L( r = 0.9991); RSD of precision,stability and reproducibility tests were no more than 3%; average recovery rate was 97.55%( RSD = 1.50%,n = 6). The contents of kinsenoside in Jin- Xian- Lian from three Anoectochilus species were in the range of 6.37% ~22.66%,and the average content was 15.00%( RSD = 26.29%). This method was quick,simple,accurate and reliable,and could be used to determine the content of kinsenoside in Jin- Xian- Lian.
