Adsorption properties of antihypertensive peptides on magnetic agarose affinity medium

  • To study on the adsorption properties of antihypertensive peptides Ile-Pro-Pro( IPP) on magnetic agarose affinity medium.The magnetic agarose microspheres were prepared with nanoprecipitation method,which were used immobilized metal affinity magnetic nanoparticles by being activated,coupled linkers and chelated nickel ions.Adsorption kinetics and isotherm of antihypertensive peptides IPP on magnetic agarose affinity medium were determined by adsorption kinetics and equilibrium adsorption experiments.The data were fitted to intraparticle diffusion model,pseudo-first-order model,pseudo-second-order model,Langmuir models and Freundlich models,respectively. Finally,the separation efficiency was analyzed by RP-HPLC on IPP that was adsorpted by magnetic agarose affinity medium from protein mixture. Results: The IPP adsorption kinetics on magnetic agarose affinity medium fitted the pseudo second-order model.The isothermal adsorption curve followed the Langmuir model well. Adsorption quantity can reach the maximum value when adsorption time was 60 min,the p H was 8.0,IPP equilibrium concentration was 20mg·mL~(-1).The antihypertensive peptides can be separated from protein mixture by magnetic agarose affinity medium. Compared with the pure solution,adsorption quantity was decreased by 2.78%,29.80%,41.30%,81.20% when the concentration rate of IPP: BSA was 5∶1,10∶1,20∶1,40∶1.Conclusion: The antihypertensive peptides can be separated quickly by magnetic agarose affinity medium from protein mixture while there was interference effects on the adsorption quantity of IPP on magnetic agarose affinity medium in the presence of BSA.
