Study on polyphenol purification by macroporous resin and inhibition activity to pancreatic amylase of polyphenol from Docyniaindica (Wall.) Dcne leaves

To propose an optimal purification process for polyphenol extracted from Docyniaindica( Wall.) Dcne leaves and investigate the inhibition of pancreatic amylase before and after purification.Macroporous resins D-101 was found more suitable to purify polyphenols by comparing their static adsorption and desorption capacities with those of two other kinds of macroporous resin. The purification process of D-101 was determined after static and dynamic experiment.When the D-101 was adsorbed with the maximum sample volume of 10 BV sample solution containing 25 mg / m L extractive at a rate of 1 m L / min,and desorpted with 8 BV of 50% ethanol at a flow rate of3 m L / min,the content of the total polyphenol could be increased from 39.7% to 91.89%.The method proved to be simple and feasible for purification of total polyphenols extracted from Docyniaindica( Wall.) Dcne leaves. After purification,the inhibitory rate of total polyphenols extracted from Docyniaindica( Wall.) Dcne leaves increased,and the inhibition rate before and after purification was higher than that of 100 mg / m L rhubarb extract on pancreatic amylase,indicating that polyphenols of Docyniaindica( Wall.) Dcne leaves had obvious inhibiting effect on pancreatic amylase,which the purification can improve.
