Cloning and sequence analysis of wild ginseng phospholipid hydroperoxide glutathione peroxidase cDNA

  • Phospholipid hydroperoxide glutathione peroxidase( PHGPx) has significant antioxidant activity,avoiding the harm by peroxidation in the cell membrane.To clone the full-length c DNA encoding PHGPx gene from Panax ginseng,the RACE technology was used in this study. The sequence analysis showed that: the full length of the cloned fragment was 490 bp.It encoded 162 amino acids and contained 3 sub-conservative domains which were specific in eukaryonic phospholipid hydroperodixe glutathione peroxidase.The encoding protein was a stable nontransmembrane hudrophilic protein. The sequence of amino acids was similar with PHGPx of citrus and castor,homologous rates of amino acids were 76% and 75%. And then,the phylogenetic tree of nine plants was constructed.
