Optimizing conditions for alkaline protease extraction of Onchidium struma mantle protein by response surface methodology

  • The effect of alkaline protease on the rate of protein extraction of Onchidium struma mantle were studied.Then the enzymolysis conditions of alkaline protease were optimized by using response surface method(RSM)on the basis of single factor as temperature,p H value,time,liquid-solid rate and enzyme amount experiments.The results showed that the optimum enzymolysis conditions were optimized as following:temperature 56 ℃,p H value 12.6,time 3.2 h,liquid-solid rate 93 m L/g respectively.Under these conditions,the predicted protein extraction rate was 18.36%,compared to the measured value 18.30%,the relative error was 0.34%. Results showed that the conditions were reasonable and feasible.
