Study on the extraction and purification of polyphenols from Sargassum fusiforme

  • Studies were carried out on the technology for separating polyphenols crude extract from Sargassum fusiforme efficiently and economically. The ethanol concentration, solid-liquid ratio, extraction time and extraction temperature were optimized by orthogonal test depending on their effects on polyphenols crude extract extracting yield. The optimized factors were ethanol concentration of 40%, solid-liquid ratio 1∶25, extraction time5 h and extraction temperature 70℃. Ten different types of macroporous resin were surveyed and selected when the adsorption capacity and desorption rate taken as indicators. The adsorption conditions of the selected macroporous resin were determined by dynamic adsorption and desorption experiment. The results showed that NKA-9 resin had the best adsorption capacity and static desorption rate for the polyphenols from Sargassum fusiforme. The adsorption capacity and desorption rates were 0.73mg/g and 91% respectively. The optimum conditions for elution were column liquid p H4, liquid column volume 300 m L, feeding rate 1m L/min;eluent concentration 70%, eluent volume 400 m L, elution rate 1m L/min.
