Optimization of microware-assisted enzyme extraction for polysaccharide from rice hull by response surface methodology

  • In order to optimize the extraction method of polysaccharide from rice hull by microware-assisted enzyme extraction technology, on the basis of single-factor experiments, cellulose addition amount, microwave powder and solid-solvent ratio were selected as influencing factors and the content of polysaccharides as response during extraction.The experiment method was designed according to Box-Behnken central composite experiment, which studied each variable and their interactions effects on the contents of polysaccharide. The quadratic multinomial model on the contents of polysaccharide was established by using SAS software. The optimum condition of extracting process was determined by means of response surface method. The results showed that the optimum extraction conditions for polysaccharide were cellulose addition amount 1.00%, solid-solvent ratio 1∶29 ( g∶mL) , microwave powder 560W and microwave time 2min.Under the optimal extraction conditions, the maximum yield of polysaccharide was 1.127%.
