A comparison of methods for the extraction of bacterial genomic DNA in rat feces

  • In order to obtain high-quality bacterial genomic DNA, and to study the diversity of the intestinal flora, improved lysozyme method, and two kinds of DNA extraction kits were used to extract bacterial genomic DNA in rat feces. These extraction techniques were compared through measuring DNA concentration and purity, and PCR-DGGE technique to find the best method for extracting bacterial genomic DNA in rat feces. Results showed that bacterial genomic DNA was extracted by improved lysozyme method had better quality and purity, and diversity of flora. It was more suitable for the subsequent intestinal microflora molecular biology research.
