Research of the solution viscosity and conformation character of polysaccharide from the mushroom sclerotia of pleurotus tuber-regium

  • Congo- red reaction and the technique of CD were used to observe morphology of polysaccharides ( SHNP) from Pleurotus tuber-regium.Inherent viscosity and the particle size distribution changes of the SHNP in different solution were studied to determine conformation of SHNP in solution.The results indicated that: the external conditions such as temperature, pH, denaturants and metal ions etc could affect SHNP to differ extent.The particle size of SHNP became bigger in the addition of NaOH.Congo- red reaction and the technique of CD showed that SHNP did not have the triple helix structure, while it was a random coil chain conformation, pH and denaturants could cause the SHNP conformation change.
