Study on optimization of enzymatic extraction of polysaccharide from base of Flammulina velutipes stipe using response surface methodology

  • Objective:Polysaccharide in base of Flammulina velutipes stipe was extracted with the assistance of enzymatic hydrolysis.Methods:Papain gave relatively higher polysaccharide yield than cellulase and pectinase, and was chosen for further study.Based on single experiment, a quadratic regression rotational combinational design involving the main three factors at three levels was employed, and data obtained were analyzed with response surface methodology.Results:Polysaccharide yield was most significantly affected by extraction temperature, followed by extraction time and pH.An optimum polysaccharide yield of 5.90%, polysaccharide obtain rate of 9.34% (on the basis of base of F.velutipes stipe weight) , polysaccharide purity of 67.3% and protein content of 5.3% (on the basis of crudue polysaccharide) were obtained when F.velutipes stipe was treated with 1.5% papain at pH5.5, 52℃ for 82min in a material-to-water ratio of 1∶20.Conclusion:The obtain rate and purity of polysaccharide of enzymatic extraction were significantly higher than of conventional hot water extraction.
