Optimization of extraction conditions for protein from pine pollen

  • In order to make full use of protein in the pollen, and to determine the basic set of pine pollen, the alkali extraction of pine pollen in protein extraction and acid precipitation were studied. The orthogonal experiment to determine the best extraction conditions of pine pollen protein solution. Barley malt roots contained 20.32%protein, 3.19% ash, 8.72% water, 2.84% fat and 24.71% reducing sugar content. Pollen protein isoelectric point of pI was 3.6, on this basis to research in solid-liquid ratio, alkali extraction time, alkali extraction temperature, alkali extraction rate for the effect of pH on pollen protein extraction. Through single factor experiment to select the optimum level, to design orthogonal experiment, to apply the range analysis and variance analysis, and reached a conclusion of the optimum extraction conditions of pine pollen protein which was the ratio of material to liquid to 1∶40 (g/mL) , alkaline extraction pH was 11, alkali extraction time was 80min, the temperature alkaline extraction for the 55℃. Under these extraction conditions, the extraction rate of pine pollen protein was 56.3%.
