Optimization of the extraction process followed by capillary zone electrophoresis for the simultaneous determination of oleanolic acid and ursolic acid in Celastrus orbiculatus Thunb.

Abstract
An optimized response surface methodology ( RSM) method and an efficient capillary zone electrophoresis (CZE) analysis method were developed for fast extraction and simultaneous determination of oleanolic acid (OA) and ursolic acid (UA) in Celastrus orbiculatus Thunb. (C.orticulatus Thunb.) . The results displayed that the optimized conditions to extract and determine OA and UA simultaneously in C.orticulatus Thunb. were 80% methanol, solvent to material ratio 20mL·g-1and extraction time 50min.Under the optimized UAE method and an efficient CZE analysis method, the contents of OA in root, stem, bark and leaf of C.orticulatus Thunb. were 0.2561, 0.6748, 0.5190, 0.1832mg·g-1. The contents of UA in root, stem, bark and leaf were 0.3720, 0.1558, 0.1097, 0.0677mg·g-1. The results showed that there were significant variations in the contents of OA and UA in different parts of C.orbiculatus Thunb, RSM was a handy extraction method with higher yield and lower solvent consumption. The CZE analysis method was validated to have good linearity, precision, reproduction and accuracy. The study provided theoretical foundation for developing medicine resource of C.orticulatus Thunb..
