Optimization of initial fermentation media of coliform group by response surface methodology

  • The initial fermentation media of coliform group was optimized by single factor investigations, Box Behnken response surface methodology based on LST fermentation media.The influences of three factors: lactose, tryptone and yeast powder on the TVCs of coliform group were studied.Finally, the appropriate medium composition was lactose 0.5g/100mL, tryptone 1.64g/100mL, yeast powder 0.29g/100mL.Under these conditions, the TVCs of coliform group was 1.025 × 107cfu/mL.The actual value was 1.033 × 107cfu/mL through verification test.The actual value had no significant difference compared with the theoretical value.
