Simultaneous analysis of chlorogenic acid and its five major metabolites compounds by high performance liquid chromatography

  • A method had been developed for the simultaneous determination of the contents of chlorogenic acid, caffeic acid, 3-hydroxyphenylpropionic acid, p-coumaric acid, ferulic acid and hippuric acid by high performance liquid chromatography.Six phenolic compounds were separated on a Agilent Eclipse XDB-C 18 (250 ×4.6mm, 5μm) column using CH 3 OH and 1% acetic acid aqueous solution as mobile phase with near gradient elution, and detected by ultraviolet detector (UV) at wavelength of 320nm and 253nm.There were good linear correlations between the concentrations and peak areas of the six phenolic compounds with the correlation coefficients of 0.9979~0.9999.The average recoveries were 86.84%~101.56% with relative standard deviations of 1.15% ~2.29%.The results suggested that this method was characteristic of rapidity, accuracy, high sensitivity.The method can be used for qualitative detection of chlorogenic acid and its metabolites.
