Optimization of derivatization conditions for the determination of 1-deoxynojirimycin by response surface methodology

  • For the purpose of establishing a high performance liquid chromatography method for the determination of 1-deoxynojirimycin (1-DNJ) , the optimal derivatization conditions were studied by response surface methodology. Based on the result of single factor experiments, the Box-Behnken design was used to investigate the effect of derivative time, temperature and the amount of derivative reagent on the recovery of 1-deoxynojirimycin. The derivative conditions were found to be 22min, 29℃ and 0.43mmol/L and the recovery was 99.4%.
