Separation and identification of rebaudioside A in stevia by high performance liquid chromatography-refractive index detector

  • A high performance liquid chromatography method for separation and determination of rebaudioside A (RA) in stevia was established. The analytical column was Waters NH 2 (250mm ×4.6mm, 5μm) , the mobile phase was acentonitrile -water (8020 , V/V) , the samples were determined by an refractive index detector detector. The calibration curves had good linearity in the range of 0.1~1.0mg/mL for RA, the detection limit for RA was 0.03mg/mL, the average recoveries of RA were 98.37% (n=6, RSD=0.88%) . The method was convenient, rapid and effective for qualitatively analysis of RA in stevia.
