Determination of tomatine in tomato and its processed products by high performance liquid chromatography

  • A method was developed for determination of tomatine in tomato and its processed products by high performance liquid chromatography (HPLC) .The tomatine in sample was extracted by methanol, cleaned up with PCX solid-phase extraction cartridge, then acetylated by acetic anhydride and determined by HPLC.The mobile phase consisted of 90% acetonitrile and 10% water.The tomatine was separated on C18 column and determined with UV detector at 205nm.The results showed:the detection limit of tomatine was 0.5mg/kg, the average recoveries were from 90.5% to 93.5%, and the relative standard deviations (RSDs) was from 1.7% to 3.2%.This method was accurate, sensitive and effective for determination of tomatine in tomato and its processed products.
