Abstract:
To improve the utilization and economic value of
Symplectoteuthis oualaniensis resource, the enzymatic hydrolysis process of
Symplectoteuthis oualaniensis tissue protein for the production of acid soluble peptides was investigated. First of all, the different types of proteases on hydrolysis of
Symlectoteuthis oualaniensis tissue protein were compared, and trypsin was determined as the preferable protease suitable for the hydrolysis process. Then, the hydrolysis process conditions were analyzed and optimized by single factor experiments and response surface methodology. Finally, the optimum process conditions were determined:protein concentration 3%, enzyme dosage 4210 U/g, pH9.23, temperature 52.3℃, reaction time 5 h. Under the optimized conditions, the yield of acid soluble peptide could reach (74.02%±0.41%). In addition, the results of amino acid composition showed that the acid soluble peptides ccould effectively maintain the amino acid composition of Symlectoteuthis oualaniensis tissue protein, 8 essential amino acids were abundant, accounting for about 38% of total amino acids. All of these results showed that the preparation process of acid soluble peptides from hydrolysis of
Symplectoteuthis oualaniensis tissue protein by protease was feasible.
