Abstract:
Objective:The extraction process of the adenosine triphosphate(camp)from JuJube was optimized,and the suitable excipients were selected to optimize the forming process of Camp granules. Methods:The extraction yield of cAMP was used as the evaluation index,and the ratio of material to liquid,extraction time,ethanol volume fraction,microwave frequency and microwave treatment time were optimized by the response surface method. The solubilities of soluble starch,lactose,dextrin and its mixture of formability,bulk density,the angle of repose,the melting rate and the hygroscopicity were taken as the indexes,and the appropriate granule excipients were selected. Then,the optimal ratio of excipient was selected by orthogonal experiment design. Concentration and particle drying temperature determine the granule molding process and process validation. Results:The optimum extraction conditions were as follows:the ratio of material to liquid was 1:15,the extraction time was 7.5 h,the microwave frequency was 270 W,the ethanol volume fraction was 30% and the microwave treatment time was 10 min. under this condition,the extraction rate of cAMP was 403.6 μg/g,optimum excipient ratio of extract:lactose as 1:4,ethanol volume fraction was 75%,the drying temperature was 50℃. Conclusion:This method was feasible,reproducible,and could produce better granules and effectively control the quality of cyclic adenosine monophosphate particles granules.
