不同产地东北林蛙卵胶膜中蛋白质和水解氨基酸含量差异比较

Comparison of protein and hydrolytic amino acids in Rana dybowskii jelly coats from different localities

  • 摘要: 比较吉林省靖宇县、敦化、蛟河山区、抚松、桦甸五个不同产地2龄和3龄东北林蛙卵胶膜中蛋白质含量、水解氨基酸种类及含量。采用考马斯亮蓝法测定林蛙卵胶膜中蛋白质含量差异;建立了异硫氰酸苯酯(Phenylisothiocyanate)作为衍生试剂的柱前衍生反相高效液相色谱法,同时分析五个产地2龄与3龄林蛙卵胶膜中21种水解氨基酸的种类与含量。结果表明,以异硫氰酸苯酯(Phenylisothiocyanate)作为衍生试剂的柱前衍生反相高效液相色谱法高效、稳定、有良好的重现性,适用于东北林蛙卵胶膜中氨基酸的测定。3年蛙龄的卵胶膜中蛋白质与氨基酸含量均高于2年蛙龄,不同产地的林蛙卵胶膜中蛋白质与氨基酸含量均有显著性差异。其中,蛟河产3龄林蛙卵胶膜中蛋白质、氨基酸含量以及必需氨基酸占氨基酸总数的比例均为最高,其蛋白质含量为(174.33±4.55)mg/g,水解氨基酸总量为(10.004±0.20)mg/g,其中必需氨基酸占比达到24.57%。抚松产2龄林蛙卵胶膜中蛋白质、氨基酸总量最低,含量分别为(147.39±1.09)mg/g、(6.414±0.10)mg/g;桦甸产2龄林蛙卵胶膜中必需氨基酸占比最低,为20.54%。蛋白质含量、氨基酸总量与必需氨基酸占比最高与最低间分别相差26.94 mg/g、3.59 mg/g和4.03%。 

     

    Abstract: To compare the protein contents, as well as the amino acids and their contents in the Rana dybowskii jelly coat from Jingyu, Dunhua, Jiaohe, Fusong, Huadian and at different ages. Protein content was determined by Coomassie method. And the type, and the method for the determination of 21 kinds of hydrolyzed amino acids in Rana dybowskii jelly coat by high performance liquid chromatography with pre-column derivatization of Phenylisothiocyanate as derivatization reagent was established.The results showed that the pre-column derivatization reversed-phase high performance liquid chromatography with Phenylisothiocyanate as a derivatizing reagent was highly efficient, stable and reproducible, and it was suitable for the determination of amino acids in Rana dybowskii jelly coat.The contents of protein and amino acids in Rana dybowskii jelly coat from three-year-old were higher than those from two-year-old. There were significant differences in protein and amino acid contents among different origins.The highest protein, amino acid contents and the ratio of essential amino acids were found in Jiaohe three-year-old. The protein content was ( 174.33 ± 4.55) mg/g; the total amount of amino acids was ( 10.004 ±0.20) mg/g; and the essential amino acids accounted for 24.57%.The lowest protein, amino acid contents were found in Husong two-year-old, the contents were ( 147.39 ± 1.09) mg/g, ( 6.414 ± 0.10) mg/g.The lowest ratio of essential amino acids produced in Huadian at the age of 2 forest frog film in 20.54%.The content of protein, the total amount of amino acids and the proportion of essential amino acids were 26.94 mg/g, 3.59 mg/g and 4.03% respectively.
