分子印迹固相萃取高效液相色谱联用检测猪肉中的磺胺类兽药残留

Determination of sulfonamides in pork by molecular imprinted solid phase extraction combined with high performance liquid chromatography

  • 摘要: 采用本体聚合法,以磺胺二甲基嘧啶作为模板分子,甲基丙烯酸作为功能单体,甲基丙烯酸羟乙酯作为亲水性功能单体,乙二醇二甲基丙烯酸酯和3-(三甲氧基甲硅烷基)丙基丙烯酸酯同时作为交联剂,偶氮二异丁腈作为引发剂,合成磺胺二甲基嘧啶分子印迹聚合物,印迹聚合物对模板分子的饱和吸附量可以达到11.37 mg/g,选择性实验中印迹聚合物对磺胺类的吸附量明显大于竞争物,对7种磺胺类药物(磺胺二甲基嘧啶、磺胺嘧啶、磺胺甲氧嗪、磺胺异噁唑、磺胺噻唑、磺胺对甲氧嘧啶、磺胺甲基嘧啶)均具有特异性吸附效果。同时建立了固相萃取与高效液相色谱联用的方法检测猪肉中痕量磺胺类药物含量,结果表明该方法对加标的7种磺胺类药物检测限为1.74.5μg/L,回收率可达73.9%85.4%,且该检测方法简单、便捷、灵敏度高。 

     

    Abstract: Hydrophilic molecularly imprinted polymers for sulfamethazine were synthesized by bulk polymerization method with sulfamethazine ( SMZ) as the template molecule, methacrylate as the functional monomer, hydroxyethyl methacrylate as hydrophilic functional monomers, ethylene glycol dimethacrylate and 3- ( trimethoxysilyl) propyl acrylate as the cross-linker, azobisisobutyronitrileas the initiator. The adsorption capacity of the imprinted polymer to the template molecule can reach11.37 mg/g. The adsorption capacity of the smectic polymer to the sulfonamides in the selective experiment was significantly larger than that of the competitor.The results showed that the hydrophilic molecularly imprinted polymers had a higher affinity and selectivity for 7 sulfonamides ( sulfamethazine, sulfadiazine, sulfamethoxazole, sulfonamidoxazole, sulfathiazole, sulfa p-methoxyprimidine, sulfamethoxypyrimidine) . At the same time, solid phase extraction coupling to high performance liquid chromatography ( HPLC) were used to detect trace sulfonamides in pork.The results showed that the detection limits of 7 kinds of sulfonamides were 1.74.5 μg/L and the recoveries were 73.9% 85.4%.The method was simple, convenient and sensitive.
