大孔吸附树脂对鱼鳞多肽的分离及吸附特性研究

摘要: 本文主要从分子极性以及热力学角度研究了大孔吸附树脂对具有血管紧张素转化酶(Angiotensin-I converting enzyme,ACE)抑制活性的鱼鳞多肽的分离及吸附作用。采用不同浓度的乙醇(20%,40%,60%,80%,v/v)对吸附鱼鳞多肽的大孔树脂进行洗脱,得到不同极性的洗脱组分,各组分之间相对分子质量的大小没有显著的差别,但各组分ACE抑制活性差异显著。当乙醇浓度从20%增加到80%时,各洗脱组分对应的IC50值分别为:0.53、0.40、0.27和0.12mg/mL。吸附热力学实验结果表明大孔吸附树脂对具有ACE抑制活性的鱼鳞多肽的吸附过程是热推动的,升高温度有利于鱼鳞肽的吸附,DA201-C大孔吸附树脂对具有ACE抑制活性的鱼鳞多肽的吸附作用力主要是疏水相互作用。Abstract: The purification and adsorption properties of fish scales polypeptides with angiotensin- I converting enzyme ( ACE) inhibitory activities treated by DA201- C macroporous adsorption resins were studied. Different eluted fractions were gotten by gradient elution of the adsorbate resins with different concentrations of ethanol ( 20%, 40%, 60%, 80%, v/v) .The relative molecular weight distribution range of different elution fractions had no significant difference, but the ACE inhibitory activity of the fish scales peptides increased. The IC50values of different fractions were 0.53, 0.40, 0.27 and 0.12mg /mL, respectively, when the ethanol concentration from 20% to80%.The results of adsorption thermodynamics experiment showed the adsorption process was endothermic, and increasing temperature was favorable to adsorption.The main force in the adsorption of fish scales polypeptides with ACE inhibitory activities on the surface of the DA201-C resins was hydrophobic.
