微波协同酶法提取蛹虫草基质多糖工艺的优化研究

Optimization of the extraction of Cordyceps Militaris polysaccharides by microwave assisted with enzyme hydrolysis

  • 摘要: 为了得到纤维素酶协同微波法从废弃的蛹虫草培养基质中提取基质多糖的最佳工艺条件,利用Box-benhnken中心组合实验设计优化基质多糖的提取工艺,建立了纤维素酶用量、酶解时间、微波功率、微波提取时间的四因素回归模型,确定了最佳提取工艺为料液比为1∶30,纤维素酶用量1650U/g,在55℃、pH5.5条件下,酶解处理44min后进行微波提取,微波功率为480W、提取时间3.5min,在最佳条件下,蛹虫草基质多糖得率为18.45%。 

     

    Abstract: To optimize the microwave-enzyme synergistic extraction process of Cordyceps militaris polysaccharides from abandoned Cordyceps militaris medium, on the base of single factor investigation, effects of cellulase content, enzymolysis time, microwave power, and microwave extract time as well as their interactions on yield of the polysaccharides were studied by response surface methodology ( RSM) analysis.The optimal extraction conditions were obtained as follows: ratio of solid- liquid 1 ∶ 30, the amount of cellulase 1650U /g, enzymolysis temperature55℃, pH5.5, enzymolysis time 44min, microwave power 480W, extraction time 3.5min.Under the optimal condition, the yield of Cordyceps militaris polysaccharides was up to 18.45%.
