稻谷芒中羟甲香豆素的鉴定及总香豆素提取工艺

摘要: 目的:确定稻谷芒中含有羟甲香豆素类成分,并研究稻谷芒中总香豆素超声波提取法的最佳工艺条件。方法:采用TLC、HPLC法对羟甲香豆素进行定性鉴别;以出膏率和总香豆素含量为参考指标,在单因素实验基础上,采用正交实验优化乙醇浓度﹑料液比、提取温度和超声时间。结果:稻谷芒含有羟甲香豆素,总香豆素的最佳提取工艺条件为乙醇浓度为55%,料液比1∶30,提取温度为70℃,超声时间为60min,功率为96W。结论:明确了香豆素类成分是稻谷芒活性物质基础,优化出总香豆素的提取工艺稳定可行,具有较高的经济价值与应用前景。Abstract: The purpose of the paper was to confirm orysan sativa containing hymecromone and to use the ultrasonic wave extraction in the total coumarin from orysan sativa trying to get the optimum extraction conditions.Hymecromone in orysan sativa was identified by TLC and HPLC methods. The extract rate and total content of coumarins as indexes, orthogonal experimental principle based on the single factor experiments was used to optimize the concentration of ethanol, the ratio of material to liquid, temperature and ultrasonic time. The results showed that hymecromone did exist in orysan sativa and the optimal extraction conditions of total coumarins of material were as follows: ethanol concentration 55%, solid-liquid ratio 1∶30, extraction temperature 70℃, extraction time 60min, power 96W. The conclusion had reached that the coumarins were mainly active ingredients in orysan sativa and the extraction process of total coumarins was stable and feasible, which had strong adaptability, high economic value and wide application prospect.
