苦菜对尼古丁中毒小鼠血清乙酰胆碱含量的影响

摘要: 探讨苦菜对尼古丁中毒小鼠血清乙酰胆碱含量的影响。通过观察小鼠中毒症状选择尼古丁溶液皮下注射剂量,测定灌胃后不同时间尼古丁中毒小鼠血清乙酰胆碱含量,确定取血时间,以灌胃后小鼠血清乙酰胆碱含量为指标,比较不同部位及不同处理的苦菜样品对尼古丁中毒小鼠的影响。结果显示,小鼠皮下注射尼古丁溶液较适宜的剂量为6mg/kg;尼古丁中毒小鼠灌胃苦菜汁45min时血清乙酰胆碱含量降低11%,与尼古丁组和空白组之间差异均显著(p<0.05),选择45min为灌胃后取血时间;尼古丁中毒小鼠灌胃苦菜根汁或叶汁45min时血清乙酰胆碱含量分别降低14%、17%,与尼古丁组和空白组之间差异均显著(p<0.05);尼古丁中毒小鼠灌胃干苦菜提取物或鲜苦菜提取物45min时血清乙酰胆碱含量分别降低13%、16%,与尼古丁组和空白组之间差异均显著(p<0.05)。结果表明,苦菜对尼古丁中毒小鼠出现的血清乙酰胆碱含量升高有明显的抑制作用,可减轻小鼠尼古丁急性中毒所产生的烟碱样症状,且有效成分存在于苦菜全株各部分,有效成分可用60%乙醇溶液提取。Abstract: To investigate the effects of Ieris chinxensis ( Thunb.) Nakai on acetylcholine content in nicotine poisoning mouse.Selected subcutaneous injection nicotine dose by observing the symptom of mouse, serum acetylcholine content was measured at different time after orally administration to select blood obtain time, serum acetylcholine content were taken as index to contrast orally administration different part or different managed Ieris chinxensis ( Thunb.) Nakai.The results showed that more appropriate subcutaneous injection nicotine dose was 6mg /kg.After orally administration Ieris chinxensis ( Thunb.) Nakai juice, serum acetylcholine content of nicotine poisoning mouse reduce 11%.Sample group had a significant difference ( p < 0.05) to nicotine group and control group at 45min, select 45min as the blood obtain time. After orally administration Ieris chinxensis ( Thunb.) Nakai rootjuice and leafjuice, serum acetylcholine content of nicotine poisoning mouse reduce 14% and 17%. Sample groups had a significant difference ( p < 0.05) to nicotine group and control group at 45min.After orally administration extractive of dry and fresh Ieris chinxensis ( Thunb.) Nakai, serum acetylcholine content of nicotine poisoning mouse reduce 13% and 16%.Sample groups had a significant difference ( p < 0.05) to nicotine group and control group at 45min.The results indicated that Ieris chinxensis ( Thunb.) Nakai cold markedly recede the acetylcholine releasing and reduce the nicotinic symptoms of mouse, the active ingredient exist in every part of the plant and could be extracted by 60% ethanol solution.
