响应面法优化东北红松针总黄酮的超声辅助乙醇提取工艺

Optimization of ultrasonic-assisted ethanol extraction of total flavonoids from Pinus koraiensis needles by response surface methodology

  • 摘要: 以小兴安岭红松松针为原料,采用超声波辅助乙醇提取红松松针总黄酮,从而达到合理利用东北红松松针资源的目的。将料液比、乙醇体积分数、超声温度,超声时间设为主要影响因素,在单因素实验基础上,以总黄酮得率为考察对象,采用Box-Behnken中心组合设计结合响应面分析法优化红松松针总黄酮提取工艺。结果表明:当料液比为1:52 g/mL、乙醇体积分数为51%、超声温度为76 ℃、时间为103 min时,其总黄酮的得率约为5.82%。工艺条件合理可靠,为工业大量生产提取提供理论依据。

     

    Abstract: The aim of this paper was to extract the total flavonoids from Xiaoxing’anling Pinus koraiensis needles by ultrasonic assisted ethanol extraction method,and to achieve the purpose of rational utilization of the Northeast Pinus koraiensis needles. The ratio of material to liquid,volume fraction of ethanol,ultrasonic temperature,ultrasonic time were set as the main influencing factors,based on the single factor experiment,the total flavonoid yield was taken as the object of study,Box-Behnken center combination design and response surface methodology were used to optimize the extraction process of total flavonoids from Pinus koraiensis needles. The results showed that when the ratio of material to liquid was 1:52 g/mL,the volume fraction of ethanol was 51%,the ultrasonic temperature was 76 ℃ and the time was 103 min,the yield of total flavonoids was about 5.82%.The process conditions are reasonable and reliable,which provides a theoretical basis for industrial production and extraction.
