桦褐孔菌多糖对糖尿病肾病小鼠肾脏的保护作用

Protective Effect of Inonotus obliquus Polysaccharides on Kidneys of Diabetic Kidney Disease Mice

  • 摘要: 为探究桦褐孔菌多糖(Inonotus obliquus polysaccharide,IOP)对糖尿病肾病小鼠肾脏的保护作用。采用链脲佐菌素(Streptozocin,STZ)诱导与高糖、高脂饲料喂养相结合的方式建立糖尿病肾病小鼠模型,将试验小鼠随机分为对照组、模型组、阳性组(盐酸二甲双胍)和桦褐孔菌多糖低、中、高剂量组。连续灌胃小鼠4周后,观察小鼠生长状态、测量小鼠体重,测定肾脏指数、肾功能指数以及肾脏抗氧化能力并对小鼠肾脏组织进行病理检测等。结果表明:桦褐孔菌多糖剂量组均能改善糖尿病肾病小鼠体重异常以及肾脏指数增高、肾脏肿大。其中高剂量组可以极显著抑制肾脏肿胀、修复肾组织损伤,提高肾脏氧化酶活力(P<0.01),抑制丙二醛产生。桦褐孔菌多糖对糖尿病肾病小鼠肾脏的保护作用与多糖抑制小鼠机体过氧化损伤、减少脂质过氧化的合成有关。

     

    Abstract: To explore the protective effect of Inonotus obliquus polysaccharide(IOP)on the kidneys of diabetic nephropathy mice,Streptozocin(STZ)induction combined with high-sugar and high-fat feed was used to establish a diabetic nephropathy mouse model. The test mice were randomly divided into a control group,a model control group,and a positive control group(metformin hydrochloride)and low,medium,and high-dose groups of Inonotus obliquus polysaccharides. After continuous intragastric administration of the mice for 4 weeks,the growth status of the mice was observed,the body weight of the mice was measured,the kidney index,renal function index,and the antioxidant capacity of the kidneys were determined and the pathological sections of the kidneys of the mice was observed. The results showed that the Inonotus obliquus polysaccharide dose group could alleviate the abnormal body weight,kidney value increased and kidney enlarged in diabetic nephropathy mice. The high-dose group could extremly significantly inhibit kidney swelling,repair kidney tissue damage(P<0.01),improve kidney oxidase activity(P<0.01),and inhibit malondialdehyde production. In conclusion,it could be concluded that the protective effect of obliquus polysaccharides on the kidneys of diabetic nephropathy mice was related to the polysaccharides inhibiting the peroxidative damage of mice and reducing the synthesis of lipid peroxidation.
