大孔树脂分离纯化熟地黄黄酮及其对小鼠运动耐力的影响

Purification of Flavonoids Extract from Radix Rehmanniae Preparata by Macroporous Resin and Its Effect on Movement Stamina of Mice

  • 摘要: 采取动态吸附与洗脱单因素实验优化大孔树脂纯化熟地黄黄酮的工艺条件,并将动物小鼠随机平均分为空白对照组(0.1 mL/g 生理盐水)、阳性对照组(0.10 mg/g 西洋参)、低(0.05 mg/g)、中(0.10 mg/g)、高剂量(0.15 mg/g)纯化产物组,连续灌胃30 d后,对各组小鼠进行负重游泳试验,通过对负重游泳时间、肌糖原、肝糖原、血清中尿素氮、乳酸等指标的测定,考察熟地黄黄酮纯化产物对小鼠运动耐力的影响。结果表明,大孔树脂纯化熟地黄黄酮的最佳提取工艺为:pH为5,质量浓度为8 mg/mL的样品溶液60 mL,以2 mL/min流速,上样至H103大孔树脂饱和吸附后,采用150 mL 80%乙醇溶液,以1 mL/min流速洗脱,产物黄酮的含量由2.52%提高至12.18%。与空白对照相比,不同剂量的纯化产物可显著延长小鼠游泳力竭时间(P<0.05,P<0.01),提高体内肝、肌糖原的储备(P<0.05,P<0.01),加快乳酸代谢(P<0.05,P<0.01),并降低血清中尿素氮的生成(P<0.05,P<0.01),表明熟地黄黄酮有助于增强机体的运动耐力。

     

    Abstract: The optimum purified technological process was confirmed on the basis of result of dynamic adsorption-desorption single-factor experiments. Blank control group(0.1 mL/g saline solution),positive control group(0.1 mg/g American ginseng)and the low,the medium and the high dosage groups of purified products(0.05,0.10,0.15 mg/g). After 30 d of continuous gavage,weight-bearing swimming tests were performed on each group of mice. The anti-fatigue effect of flavonoids of Radix Rehmanniae Preparata was investigated by measuring the weight-bearing swimming time,muscle glycogen(MG),hepatic glycogen(HG),serum urea nitrogen(BUN),lactic acid(LD).The results showed that the best purification process conditions were as follows:the initial concentration in sample solution was 8 mg/mL,loading flow rate was 2 mL/min,loading sample volume was 60 mL,the pH value of sample solution was 5,and the ethanol concentration was 80%(v/v),the eluent volume was 150 mL with flow rate of 1 mL/min. After purification,the purity of total flavonoids in product was increased from 2.25% to 12.18%. Compared with the blank control group,the groups of flavonoid of Radix Rehmanniae Preparata at all doses significantly prolonged weight-loaded swimming time of mice(P<0.05,P<0.01),obviously improved storage capacity of hepatic glycogen and muscle glycogen in vivo(P<0.05,P<0.01),and accelerated lactic acid metabolism(P<0.05,P<0.01),decreased urea nitrogen creation in serum after sport(P<0.05,P<0.01). Therefore,flavonoid of Radix Rehmanniae Preparata could enhanced sporting stamina in body.
