黑菊芋水提液对便秘小鼠润肠通便作用的研究

Laxative and Regulating Function of Black Jerusalem Artichoke Water Extract on Constipation Mice

  • 摘要: 目的:研究黑菊芋水提液对便秘小鼠肠蠕动和排便功能的影响。方法:使用复方地芬诺酯建立小鼠便秘模型,将KM小鼠随机分成9组,分别为空白对照组、模型对照组、阳性对照组、鲜菊芋低、中、高剂量组和黑菊芋低、中、高剂量组,低、中、高剂量组对应的给药量分别为2.5、5.0和10.0 g/kg bw,检测小鼠的体重变化、小肠推进运动、排便情况及粪便中短链脂肪酸含量等指标。结果:与对照组相比,模型组小鼠的首粒黑便时间显著推迟(P<0.05),肠墨汁推进率,6 h内排便粒数及排便重量显著降低(P<0.05),粪便中短链脂肪酸总量显著降低(P<0.01),说明便秘模型建立成功。与模型组相比,5.0和10.0 g/kg bw黑菊芋水提液能够显著缩短便秘小鼠首粒黑便时间(分别提前24.57%和36.56%),增加小鼠黑便粒数(分别增加67.64%和85.29%)和黑便质量(分别增加87.31%和67.26%),增强小鼠小肠墨汁推进率(分别增加70.59%、101.02%),促进小鼠肠道代谢产生乙酸、丙酸和丁酸,提高肠道中短链脂肪酸的总量。结论:5.0和10.0 g/kg bw黑菊芋水提液可以有效促进便秘小鼠的肠道蠕动,缓解便秘,具有润肠通便的功能,本研究为黑菊芋的保健功效提供了依据。

     

    Abstract: Objective:To study the laxative and regulae function of black jerusalem artichoke water extract on constipation mice induced by compound diphenoxylate. Mice were randomly divided into 9 groups,namely control check treatment(CK),model control treatment(MC),active control treatment(AC),low,medium and high dose treatment of raw jerusalem artichoke,and low,medium and high dose treatment of black jerusalem artichoke,the doses corresponding to the low,medium and high dose treatment were 2.5,5.0 and 10.0 g/kg bw,respectively. Then the changes of body weight,intestinal transit,defecation situation,and the contents of short-chain fatty acids in feces of mice were measured. Results:The results showed that compared with mice in CK,the time of first melena of mice in MC was significantly delayed(P<0.05),the ink propelling rate,the number and mass of defecation particles within six hours were significantly reduced(P<0.05),and the total SCFAs in feces was significantly reduced(P<0.01),indicating that the constipation model was successfully established. In comparison with mice in MC,the mice fed on black jerusalem artichoke extract of 5.0 and 10.0 g/kg bw could significantly shorten the time of first melena by 24.57% and 36.56%,increase the number of melena by 67.64% and 85.29% and the quality of melena by 87.31% and 67.26%,increase the ink propelling rate by 70.59% and 101.02%,respectively.Furthermore,the amount of short-chain fatty acids in the intestinal,such as acetic acid,propionic acid and butyric acid were increased. Conclusion:5.0 and 10.0 g/kg bw black jerusalem artichoke extract can effectively promote constipated mice intestinal wriggling,improve intestinal environment and alleviate constipation,and has the function of laxative and regulating.This study provides data support for the health benefits of black jerusalem artichoke.
