一种酸枣仁提取物、茶氨酸、γ-氨基丁酸配方产品改善睡眠作用研究

Sleep Improvement Effects of Semen Ziziphi Spinosae Extracts,Theanine and γ-Aminobutyric Acid Formula Products

  • 摘要: 目的:探讨一种酸枣仁提取物、茶氨酸、γ-氨基丁酸配方产品改善睡眠作用。方法:180只健康雄性Balb-c小鼠按体重随机分为三个大组,每个大组60只小鼠分别随机分为空白对照组和低、中、高3个剂量组,每组15只。酸枣仁提取物、茶叶茶氨酸和GABA配方产品按照低、中、高3个剂量(95、190、285 mg/kg·bw)通过直接睡眠、延长戊巴比妥钠睡眠时间、戊巴比妥钠阈下催眠剂量和巴比妥钠睡眠潜伏期实验,研究酸枣仁提取物、茶氨酸、γ-氨基丁酸复合配方对小鼠睡眠的影响。结果:酸枣仁提取物、茶氨酸与γ-氨基丁酸配方产品无直接催眠作用。与对照组相比,低、中、高剂量组小鼠戊巴比妥钠睡眠时间分别延长14.53%、17.12%、23.80%,且差异均具有统计学意义(P<0.05)。各剂量组小鼠30 min内小鼠阈下剂量实验均为阳性,小鼠睡眠发生率增加,差异具有统计学意义(P<0.05)。各剂量组小鼠巴比妥钠睡眠潜伏期时间分别缩短12.48%、24.19%、24.07%,差异有统计学意义(P<0.05)。结论:酸枣仁提取物、茶氨酸、γ氨基丁酸配方产品具有缩短小鼠睡眠潜伏期,延长小鼠睡眠时间,同时增加小鼠入睡发生率的作用,具有改善睡眠的功效。

     

    Abstract: Objective:To research the sleep improvement of Semen Ziziphi Spinosae,theanine and γ-aminobutyric acid formula products in mice. Methods:180 healthy male Balb-c mice were divided randomly into three groups. Each group was divided into four groups:One control group and three treatment group with equal size. Semen Ziziphi Spinosae,theanine and γ-aminobutyric acid were mixed in proportion and fed to mice at 95,190,285 mg/kg·bw respectively for 30 d by oral administration. Then direct sleep test,sleep extending test with sodium pentobarbital,hypnosis test under subthreshold dose of sodium pentobarbital and incubation period test with barbital sodium were conducted respectively. Results:The experimental samples had no direct hypnotic effect. The sleep time of mice injected with pentobarbital sodium in the low,medium and high dose group was longer by 14.53%,17.12%,23.80% compared with the model group(P<0.05). The rate of sleeping increased under the threshold hypnogenesis dosage of pentobarbital sodium in medium and high dose group(P<0.05). The sleeping delitescence induced by tobarbital sodium significant decreased by 12.48%,24.19%,24.07% compared with the model group(P<0.05). Conclusion:The formula consists of Semen Ziziphi Spinosae,theanine and γ-aminobutyric acid can shorten the sleep incubation period,prolong the sleep time and increase the sleeping rate of mice. According the results,the formula has the sleep improvement effect in mice.
