白苏叶乙醇提取物体外抗氧化活性评价及相关性分析

Evaluation of Antioxidant Activities in Vitro of Ethanol Extract from Perilla frutescens(L.)Britt Leaves and Correlation Analysis

  • 摘要: 利用六种不同的评价抗氧化活性方法,包括细胞内活性氧清除能力(CAA)、总抗氧化活性(TAA)、清除超氧阴离子自由基(SSAR)、清除羟自由基(SHR)、清除二苯基苦基苯肼自由基(SDR)活性以及抑制卵黄脂蛋白脂质过氧化(ILLP)对白苏叶乙醇提取物(WPEE)体外抗氧化活性及清除自由基能力进行了评价,并与抗坏血酸(VC)的抗氧化活性进行了比较。结果表明,六种评价方法均证实WPEE有明显的抗氧化活性,其中TAA、SSAR和SDR活性顺序均为:VC > F2 > WPEE > F1(F1和F2为WPEE的组分1和2),但SHR及ILLP活性顺序为F2 > WPEE > VC > F1。此外,WPEE在浓度1~500 μg/mL范围内,对RAW264.7细胞是无毒性的,且具有良好的活性氧(ROS)清除能力。皮尔森相关性分析显示,六种评价抗氧化活性的方法均具有局限性,其评价的是抗氧化活性的不同方面;但除了ILLP与SSAR几乎没有相关性外,其他4种评价抗氧化活性的方法具有很好的相关性;抗氧化活性大小与WPEE的浓度正相关,说明WPEE是有重要应用前景的广谱型天然抗氧化剂。

     

    Abstract: Six different kinds of methods to evaluate the antioxidant activities,including intracellular reactive oxygen species scavenging abilities(CAA),total antioxidant activities(TAA),scavenging superoxide anion radical(SSAR),scavenging hydroxyl radical(SHR),scavenging 1,1-diphenyl-2-picrylhydrazyl radical(SDR)and inhibition of lipoprotein lipid peroxidation(ILLP)were used to evaluate the antioxidant activities in vitro and free radical scavenging abilities of Perilla fruescens Britt leaves,ethanol extract(WPEE),which were compared with antioxidant activity of ascorbic acid(VC).The results showed that six kinds of methods all indicated that WPEE had significant antioxidant activities. The order of TAA,SSAR and SDR was VC > F2 > WPEE > F1(F1 and F2 were the main components 1 and 2 of WPEE),while the order of SHR and ILLP was F2 > WPEE > VC > F1.In addition,WPEE was non-cytotoxic to RAW264.7 cells and had a good scavenging ability of reactive oxygen species(ROS)within the concentration range of 1~500 μg/mL. Pearson correlation analysis showed that the six kinds of methods of evaluating antioxidant activity were all limited,which evaluated different aspects of antioxidant activity. Except for ILLP and SSAR,the other four kinds of methods to evaluate antioxidant activity had a good correlation with each other. The antioxidant activity was positively correlated with the concentration of WPEE,indicating that WPEE was a broad-spectrum natural antioxidant with important application prospects.
